Genome-Wide Identification of the BvCBL Genes in Sugar Beet (Beta vulgaris L.) and Their Expression Under Salt and Drought Conditions

英文摘要Salinity and drought are two major abiotic stresses limiting the growth and development of plants and the productivity and quality of crops worldwide. Calcium (Ca2+), as a ubiquitous second messenger, mediates the response of plants to salt and drought stresses. The calcineurin B-like protein (CBL), as an essential Ca2+ sensor, interact with CBL-interacting protein kinase (CIPK) to constitute a CBLs-CIPKs signaling network, which enables information integration and physiological coordination to cope with extracellular abiotic stresses. To understand the biological function of CBLs in sugar beet (Beta vulgaris L.) during abiotic stress tolerance, in this study, the BvCBL genes were identified firstly by bioinformatics methods, and their expression patterns under NaCl and sorbitol treatments were investigated. The results showed that a total of seven BvCBL genes were distinguished and characterized in the genome of sugar beet. The BvCBL genes contained 7 similar to 10 exons and 6 similar to 9 introns. The number of amino acids was 213 similar to 289 aa, the molecular weight was 24.56 similar to 33.25 kDa, and the isoelectric point was 4.47 similar to 5.05. The BvCBL genes were distributed on four of nine chromosomes and can be divided into four distinct groups, namely Group I, II, III, and IV, according to the sequences homology. Among the candidate BvCBLs, BvCBL6 had two EF-hand motifs, while the others contained four ones. The palmitoylated sites were found in all the BvCBL genes, while myristoylation sites were observed only in three genes, BvCBL1, -2, and -5. The promoter of the BvCBL genes mainly contained eight stress responsive elements and nine hormone responsive elements. The expression pattern of the BvCBL genes were further analyzed in sugar beet under NaCl and sorbitol treatments, and found that 50 mM NaCl significantly induced the expression levels of the BvCBL genes in both shoots and roots, while different concentrations of sorbitol significantly increased the expression abundance of genes in roots but not in shoots. Furthermore, the BvCBL1 gene was overexpressed in Arabidopsis thaliana, and found that the overexpression of BvCBL1 significantly enhanced salt tolerance in transgenic plants compared to wild-type plants. The results of this study could provide not only a theoretical support for further research on the biological function of the CBL genes under abiotic stress, but also a basis for the selection and breeding of salt- and drought-tolerant varieties of sugar crops.
