Combined Effect of Ultrasound and Phanerochaete chrysosporium Pretreatments of Distillers Grains for Fermentable Sugars by Enzymatic Hydrolysis

英文摘要Distillers grains (DG), mainly composed of lignin, cellulose and hemicellulose, is one of the most abundant food processing byproducts producing from white spirit brewage in China. To produce more fermentable sugars from DG by enzymatic hydrolysis, the ultrasound (U) and the combination of biological pretreatment (B) by Phanerochaete chrysosporium were evaluated under different conditions. Moreover, the effects of ultrasound pretreatment parameters on the yield of reducing sugar (YRS) were optimized by the response surface methodology (RSM), and the optimum pretreatment conditions were selected. The structural characteristics changes were studied by scanning electron microscopy (SEM), X-diffraction (XRD) and fourier-transform infrared spectrometry (FTIR). The optimal ultrasound pretreatment condition was identified as ultrasound power 420 W, time 39 min, temperature 40 degrees C, and solid-liquid ratio 1.22 (g/mL). Under this condition, YRS was reached to 68.73%. The combining pretreatment results showed that the combinations led to a significant increase in YRS as compared with that from any single pretreatment. YRS was reached up to 74.65% when using ultrasound firstly (U + B). The SEM showed significant structural changes in U + B pretreated DG, the surface of U + B became increasingly coarse, the gaps between cellulose fibers were visible, and many pores were developed. Moreover, X-ray diffractometry and FTIR analysis revealed that four pretreatments can weaken the recalcitrance by breaking the lignin-carbohydrate bonds and removing amorphous hemicelluloses and obstinate component lignin to increase the crystalinity of cellulose. However, the cellulose crystallinity of pretreated DG in case of U+ B pretreatment was lower than that of other pretreatment methods, and the strength of characteristic peaks for corresponding typical functional groups of each component are changed compared with the feedstock. This suggests that the combination of ultrasound and Phanerochaete chrysosporium pretreatment is effective in improving the enzymatic hydrolysis efficiency of DG.
