英文摘要Protein fouling of membranes is an obstacle to the wide application of microfiltration in biotechnology. This thesis investigated three aspects of protein fouling of membranes, including the cause of membrane fouling, the formation of aggregate in solutions, and the effect of concentration of aggregates on flux of microfiltration. A novel method for determining the amount of proteins on membranes was developed, which was precise, stable and repeatable. The method can be applied to commonly used membranes such as mixed esters of cellulose nitrate and acetate membranes, Durapore GVWP membranes, and track-etched polycarbonate membranes. Adsorption and deposition of proteins on membranes was observed under both static and filtrating conditions. It was shown that adsorption and deposition of proteins on membranes occurred simultaneously in microfiltration. Adsorption of proteins would be saturated soon after the beginning of microfiltration. The amount of proteins that deposited on membranes was proportional to the volume filtrated. The result also showed that the existence of aggregates in solution was the major factor that caused membrane fouling. Formation of aggregates in protein solutions was studied. It was shown that the pumping process was the major cause for the formation of aggregates. High temperature, high concentration of proteins, and a pH value near isoelectric point can accelerate the formation of aggregates. The results also showed that the amount of aggregates formed in solutions was linearly proportional to the average shear rate generated by pumping. Flux decline in microfiltration was studied, and a mathematical model describing flux was constructed. The model was in good agreement with the experimental data. Parameters of the model were directly related to the concentration of aggregates and the pumping treatment of the solution. The size of aggregates can be calculated with the values of the model parameters, and their size was similar to that reported in the literature.
