中文摘要固态发酵以其自身的特点、优势以及其在许多领域中的广泛应用潜力正受到越来越多的重视.但在其推广和实际应用过程中,还需要克服和解决固态发酵过程中的传质、传热难,放大过程中纯种培养难等问题.该文采用固态发酵生产碱性蛋白酶,探讨固态发酵过程中存在的问题,提出解决的办法,为固态发酵推广应用提供一定的参考依据.该文首先研究麸皮固态发酵碱性蛋白酶的可行性.然后利用惰性载体吸附固态发酵这一新型固态方式,进行碱性蛋白酶发酵的尝试.
英文摘要Much attention has been paid to solid-state fermentation (SSF) recently because of its characteristics, advantages, and its potential applications in many fields. However, there are also many difficulties, such as the low efficient transfers of oxygen and heat during SSF and difficulties in pure cultivation during scale-up of SSF. In this paper, SSF was adopted to produce alkaline protease. And some measures to were advanced to resolve the difficulties during SSF, which might be helpful to the practical utilization of SSF. Firstly, SSF was applied to produce alkaline protease using wheat bran as the only substrate. As a result, the yield reached 210U/g wheat bran and the wheat bran could be recycled. In order to improve the transfer process in SSF, air pressure pulsation SSF as a technical platform was adopted and the yield got to 430U/g wheat bran. However, the yields were both very low. Secondly, SSF using inert carrier as solid support, as a novel SSF, was applied to produce alkaline protease. When 15.0ml of liquid medium with CaCb at 15.0mM concentration was adsorbed by 4.0g of polyurethane foam, at temperature of 32°C, initial pH of 9.0 and inoculum's size of 1.0%(v/v), after cultivation of 24hours, the yield got to 2180U/ml, which was very comparable to that of submerged fermentation (SmF). During the SSF using inert solid carrier as support, the inert solid carrier and the nutrient medium were self-existent, which might be helpful to the realization of continuous SSF. This paper came up with a continuous SSF using inert carrier as solid support to produce alkaline protease. When the flow rate of the medium was 0.05ml/min, and the flow rate of the air was lOml/min, the yield got to 1420U/ml. However, compared with that of SmF, the yield was lower. More work should still be done to improve the effect of this kind of continuous SSF.
