英文摘要 | Methamphetamine (METH) priming is critically involved in activating longlasting relapse and compulsive drug seeking. The purpose of the preset study was to establish a rat model of METH relapse behavior induced by METH priming. Male Sprague Dawley rats were trained to stably self-administer METH (0.06 mg/kg per infusion) under a fixed ratio 1 schedule for 8 consecutive days. After 2 weeks of withdrawal, the rats were returned to their training chambers for 7 h relapse testing induced by contextual (chambers) and METH priming, respectively. We found that a total of 75% of rats has established stable METH intravenous self-administration after 8 days of training. After 2 weeks of withdrawal, METH seeking elicited by contextual cues (self-administration chamber) or METH priming was measured in two consecutive 7 h test phases. During both test phases, the animals exhibited extinguished responding and the responding was most prominent during the first contextual sessions. Finally, the animals showed increased responding of nose poking on the METH priming during the period of reinstatement. The results suggested that self-administration responding could be reinitiated by METH-induced after a period of abstinence. The procedure used in this model may be useful for the study of the underlying mechanism of drug-induced reinstatement. |
