中文摘要 | triacylglycerides (tags) are a large class of complex neutral lipids that naturally occur in both plants and animals. these compounds are important to organisms as they are the main energy resource for biological activities, but when the tags were excessively accumulated they could cause metabolic syndromes such as obesity and diabetes. this needs us to better understand the individual tag molecules or profile their changes in organism and firstly we should isolate these compounds. a direct and effective way to do this is to perform chromatography. however, tag species are very complex since they are composed by the seterification of a glycerol backbone with three fatty acids, any change of the position or chain length or the number or position of the double bonds of the fatty acids changed the tag molecule. thus the separation of tag puts more demands on the efficiency and peak capacity of the separation approach and an elegant option for this is the comprehensive two dimensional liquid chromatography. in the present work, we used an on-line comprehensive silver-ion liquid chromatography (silver-ion lc) ×reversed-phase liquid chromatography (rp lc) system to analyze triacylglyceride compounds. first dimensional separation was performed on a micro bore silver-ion modified column employing the commonly used hexane-based mobile phase. optimization was focused on the second dimension, and a series of c18 columns were assessed. a wide bore column packed with 1.5 μm particles was selected as the second dimension column to reduce the negative effect caused by the large volume and strong solvent injection in the second dimension. the system coupled with mass spectrometry was applied to the analysis of the mouse liver extract. two mouse livers, a yin-deficiency model and its control, were studied and totally 52 tags were obtained. the identification was based on the tags’ retention behaviors on the comprehensive two-dimensional lc system and their apci ms fragments. of the 52 identified tag, 28 were shared by the two samples, leaving 6 from the model and 18 from the control respectively. this demonstrated that the discrimination between the two types of sample was feasible and so was the application of this separation technique in |
