英文摘要在pH 1.4的Clark-Lubs 缓冲介质中,固绿FCF与血清蛋白质作用在室温下能迅速结合形成复合物,其最大吸收波长为660 nm,比固绿FCF本身红移了36 nm.用分光光度法研究了该结合反应的最佳条件,并在此基础上建立了测定蛋白质的新方法.在660 nm处各蛋白质浓度至少在5~70 mg/L的范围内与吸光度成正比,对测定牛血清白蛋白(BSA)、人血清白蛋白(HSA)及球蛋白(IgG)的表观摩尔吸光系数ε660和桑德尔灵敏度s分别为7.87×105、9.52×105、1.60×106 L*mol-1*cm-1和0.083、0.072、0.093 μg*cm-2.除阴、阳离子表面活性剂外,其余大部分物质不干扰蛋白质的测定.所拟方法具有简便、快速、选择性好、灵敏度高等特点,应用于尿液、人血清及含乳饮料中总蛋白的测定,结果与考马斯亮蓝G-250法基本一致.; In the Clark-Lubs medium with pH 1.4, fast green FCF can bind rapidly to form a complex with serum protein at room temperature, which has a maximum absorption wavelength at 660 nm, and exhibits a bathochromic shift of 36 nm compared with the maximum absorption wavelength of fast green FCF. The optimum reaction conditions were investigated, and a new method for the determination of proteins was developed based on this binding reaction. The absorbance of the binding system at 660 nm is proportional to the concentration of some proteins in the range of 5similar to70 mg/L with a molar absorptivity of 7.87x10(5), 9.52x10(5), 1.60x10(6) L.mol(-1).cm(-1) and Sandell sensitivity of 0.083, 0.072, 0.093 mug/cm(-2) for the determination of bovine serum albumin (BSA), human serum albumin (HAS) and human immunoglobulin G (IgG), respectively. A lot of the foreign substances do not interfere with the determination of proteins except for the anionic surfactant and cationic surfactant. The proposed method, which is rapid, stable, and of good selectivity and high sensitivity, has been used for the determination of the total proteins in urine and human serum samples as well as milk drink samples.
