英文摘要 | Fructose-1,6Diphosphate is an energy metabolism intermediate, and in recent years, it has shown many important functions in cells. So, it is necessary to develop methods for the quantification of Fructose-1,6Diphosphate in cells. A high-performance liquid chromatography (HPLC) method is developed in this study for analysis of the intracellular intermediate. This study provides a way to have an insight into the content of Fructose-1,6-Diphosphate in Human renal epithelial cell line 293T cells. An Angela Venusil MP C18 column and isocratic elution are adopted. Mobile phase A: 50 mM ammonium hydroxide, mobile phase B: 100% methanol. Isocratic elution and 5% methanol, the linear range is 0.1 similar to 50. M. LOD (S/N. 3) is 0.0001. M, and LOQ (S/N. 10) is 0.01. M. The fructose-1,6diphosphate in human renal epithelial cell line 293T cells is within the liner range. The developed method in this study is applied to determine fructose-1,6-diphosphate in human renal epithelial cell line 293T cells successfully. |
