A self-assembling method to prepare lactoferrin nanoparticles for photosensitizer delivery

英文摘要Protein-based nanoparticles hold great application in both preclinical and clinical due to their high biocompatibility and biodegradability. The present work deals with the design and process optimization for preparation of lactoferrin nanoparticles as carrier for delivery of photosensitizer to treat tumor. Lactoferrin (LF) and photosensitizer (ICG) were mixed with 50 mM GSH at 37 °C for 1 h to break disul fide bonds. Then, 2 mL of the ethanol was added into the solution to precipitate the LF-ICGNPs and kept under stirring for 30 min. The suspension was dialyzed. Using this method, LF-ICGNPs could be readily obtained. The hydrodynamic size of LF-ICGNPs was 130 nm with polydispersity of 0.100-0.150. The absorption spectra displayed that ICG was encapsulated in LF-ICGNPs. ICG absorbed NIR light and converted it into cytotoxic heat for tumor. Therefore, LF-ICGNPs can provide a noninvasive assistant tool for cancer phototherapy.
