Hemoglobin-loaded albumin nanoparticles as targeted oxygen carrier for enhanced cancer photodynamic therapy

英文摘要In photodynamic therapy (PDT), particular attention has been focused on oxygen supply of solid tumors, which is a crucial role in determining the generation of cytotoxic reactive oxygen species (ROS). However, native tumor hypoxia is a major barrier for potent PDT. Safe and efficient oxygen supplying strategies are desirable to alter the hypoxic microenvironment of tumor. Herein, we reported chlorin e6 and hemoglobin loaded albumin nanoparticles with specific-targeting and oxygen-carrying capacity, to achieve more efficient PDT. By cross-linking with glutaraldehyde and self-assembly, the natural proteins (albumin and hemoglobin) formed 35 nm spherical nanoparticles. Owing to the targeting nature of albumin to cancer cells, the nanoparticles co-loading chlorin e6 delivered abundant oxygen and photosensitizer synchronously into tumor, which was testified by fluorescent imaging in vivo. Consequently, the tumor hypoxia was radically changed. Upon exposure to 660 nm laser, the remote-triggered photosensitizer generated massive ROS with sufficient oxygen supply. ROS triggered the irreversible oxidative damage of xenograft tumor resulting in complete suppression. The oxygen content in tumor was real-timely monitored by photoacoustic imaging, which could precisely guide the enhanced PDT. Such versatile theranostic nanoplatform could serve as a safe and efficient strategy of oxygen supply to break tumor hypoxia for effective cancer therapy.
