REGULATION OF TENDON-DERIVED STEM CELLS DIFFERENTIAITON BY THE DEGENERATIVE TENDON MICROENVIRONMENT

英文摘要Tendon-derived stem cells (TDSCs) were regarded as culprit for degenerative tendinopathy. However, the differentiation of TDSCs regulated by the degenerative microenvironment of tendinopathy has remained unrevealed. In this study, the effects of the degenerative microenvironment on the morphology, proliferation and differentiation of TDSCs were investigated. The degenerative tendons were obtained by culturing of rat Achilles tendon for 4 weeks ex vivo. Normal and degenerative rat Achilles tendons were longitudinally slicedlwith the thickness of 100 m and then treated with repetitive freeze/thaw procedure with nuclease. TDSCs, which were isolated from rat Achilles tendons, were cultured on the normal and degenerative tendon slices for 2 weeks. Immunofluorescence, CCK8 assay were used to show the morphology and proliferation of TDSCs. The expression of COL1, SCX, THBS4, TNMD, CLO2. SOX9, Osterix, RUNX2, and ALP were evaluated by qRT-PCR. TDSCs were elongated and aligned along the collagen fibers on normal decellularized tendon slices (DTSs) (Fig. 1A), however they were more spread on degenerative DTSs (Fig. 1B). TDSCs had higher proliferative ability compared to that on degenerative DTSs (Fig. 1C). Furthermore, tenocyte related markers COL1, SCX, THBS4 and TNMD were highly expressed on normal DTSs than degenerative DTSs, especially COL1 and TNMD. However, TDSCs on degenerative DTSs had higher expression of COL2, SOX9, and RUNX2, ALP respectively compared with that on normal DTSs (Fig. 2). TDSCs had different morphology, proliferation rate and gene expression pattern on normal and degenerative DTSs. These results indicated that in the degenerative microenvironment of tendinopathy, TDSCs tended to differentiate toward chondrogenic and osteogenic linage instead of tenogenic linage, which could lead to calcifying or even ossifying tendinopathy.
