中文摘要intracellular lipid accumulation is a common biological process for some eukaryotic microorganism under specific growth conditions, yet study on this phenomenon at an ‘-omics’ level remains rare. in this study we induced lipid accumulation by the oleaginous yeast rhodosporidium toruloides and lipomyces starkeyi by transferring cells into a nitrogen limited medium and thereafter an online multidimensional µrplc/ms/ms method was performed. 46 proteins were notably changed in r. toruloides and 95 proteins in l. starkeyi when cells were shifted from the nutrient-rich medium to the nitrogen-deficient medium for lipid production upon semi-quantitative analysis of the dataset. gene ontology annotation revealed that most down-regulated proteins are related to the tricarboxylic acid cycle, lipid metabolism, carbohydrate metabolism and glycolysis; whereas the up-regulated proteins related to proteolysis and peptidelysis. further explanation of the results were provided in regarding to cellular responses to nutrient availability under specified culture conditions as well as the biochemistry of lipid accumulation. in addition, a sequential protein extraction method was developed for the oleaginous yeast such as r. toruloides and l. starkeyi. this work presented valuable information for further exploration of the molecular mechanism of cellular lipid metabolism and future strain engineering study.
