High sensitive protein detection with in-situ fluorescence derivatization

中文摘要in the post-genome sequencing era, high sensitive protein analysis has been paid much attention. although native fluorescence emitted by aromatic amino acid residues is an effective tool for proteins detection, not all proteins could generate intensive native fluorescence signal. thus, various fluorescence reagents have been synthesized and applied for protein derivatization. however, the derivatization of low abundance proteins is still challenging. taking the traditional fluorescence reagent, fitc, as an example, the limit of detection for proteins is similar to that of uv detection. in current work, a novel in-situ fluorescence derivatization method based on hollow fiber membrane interface was developed to achieve high efficient derivatization and high sensitive detection of proteins, by which proteins were firstly concentrated in hollow fiber membrane, and then in situ derivatized by submerging the membrane into fluorescence buffer. with such a method, the high sensitive detection of proteins with pm concentration were achieved by hplc with a fluorescence detector. furthermore, the derivatization of a fraction of e. coli whole cell lysate were achieved, and high sensitive detection was obtained compared to that obtained by uv detection and conventional in-solution fluorescence derivatization.
