中文摘要a culture-independent nested pcr approach was employed to investigate the phylogenetic diversity of actinobacteria communities associated with two marine sponges hymeniacidon perleve and sponge sp. (stach et al., 2003). the phylogenetic affiliation of sponge-derived actinobacteria was assessed by restriction fragment length polymorphism (rflp) analysis (zhang et al., 2006) and 16s rdna sequencing of cloned dna fragments. of 196 positive clones screened by rflp, a total of 46 representative cloneswere selected for sequencing. phylogenetic analysis of these clones revealed representatives of 10 actinobacteria genera including acidimicrobium, gorynebacterium, propionibacterium, gordonia, micrococcus, microbacterium, streptomyces, mycobacterium, cellulosimicrobium sporichthya, and unidentified actinobacteria clones. acidimicrobium was the most abundant group in h. perleve clone library, representing 33% of the clones, with the second abundant group being sporichthya, representing 20% of the clones. there was a high predominance of mycobacteria group in sponge sp. clone library, which accounted for 70% of all clones. the second abundant group was acidimicrobium, representing 24%. these results indicated the presence of a high diversity of actinobacteria phylotypes within the sponges h. perleve and sponge sp., belonging to 11 main groups. actinobacteria community grouped within subclass acidimicrobidae was found in both clone libraries, which was closely affiliated with previous reports (taylor et al., 2007). the information obtained by the cultural-independentmethod will be useful for designing isolation and cultivation protocols of unculturable actinobacteria.
