中文摘要objective: to gain further insight into taxol biosynthesis relevant to improving production of this anticancer drug. methods: a kinetic model representing the pathway for biosynthesis of taxol has been developed. the kinetic properties of the reactions have been modeled according to available data in literature. the model is capable of describing the flux through the biosynthetic pathway. using the kinetic model, we observed the effects of enzymes on taxol generation using metabolic control analysis (mca). results: the flux control coefficients c (flux, enzyme) of last flux for each enzyme are calculated by the model. the values of c (j12, e6), c (j12, e7), c (j12, e11) and c (j12, e12) are approximately 0.16,0.15,0.145 and 0.489 separately. other flux control coefficients values are almost equal to zero. conclusion: the determined flux control coefficients suggested that, the flux is mainly controlled by the formation of the baccatin iii, the assembly of the c13 side chain and the steps in which the 10-deacetyl-2-debenzoylbaccatin iii is formed; the enzymes that control these reactions are found to be key factors for taxol biosynthesis. the model applied in the present work is still incomplete and will be revised further to better give results and serve its purpose properly.
