Estrogen-Related Receptor α Regulates Hepatic VLDL Secretion | |

英文摘要 | The orphan nuclear receptor estrogen-related receptor alpha (ERRα) is a key metabolic regulator, including responsible for lipid biosynthesis and fatty acid oxidation. However, it is currently unknown whether ERRαis required for the control of lipid transport in liver. Here, we demonstrate that ERRα influences hepatic very low-density lipoproteins (VLDLs) secretion by regulating the expression of VLDL-related genes. Treatments of ERRα inverse agonist compound 29 (C29) or XCT790 significantly decreased apolipoprotein B (ApoB), microsomal triglyceride-transfer protein (MTP) and phospholipase A2 G12B (PLA2G12B) expression in HepG2 cells. On the contrary, overexpression of ERRα using adenovirus enhanced the expression levels of these genes. Consistently, we found elevated lipid accumulation by treatments of inverse agonist;whereas, overexpression of ERRα promotes intracellular lipids clearance in primary hepatocytes. By using a high-fat-diet (HFD) mouse model, weobserved that pharmacological inhibition ofERRα activity leads to higher levels of hepatic triglycerides (TG) which accompanied with lower plasma TG.Furthermore, we demonstrated that hepatic VLDL-TG secretion rate as well as the number and size of VLDL particles reduced significantly in comparison with control groups. In addition, hepatic expression levels of ApoB, MTP and PLA2G12B were suppressed by C29 treatment. Collectively, these data unravel a previously unappreciated role of ERRα as a positive regulator for TG-enriched VLDL secretion. |
