中文摘要通过反相高效液相色谱(HPLC)方法,分析了从陕西、四川、贵州、云南、甘肃、安徽、湖南、河南等19处不同产地获取的天麻药材样品在不同炮制方法下天麻素的含量,并进行了比较分析,以期为天麻药材的质量控制提供依据,为天麻的炮制提供科学的方法。色谱柱为 Shim-pack PREP-ODS(H)KIT C18柱(5μm,250×4.6mm),流动相为甲醇-磷酸盐溶液-水(1.5: 3:95.5),流速为1.0mL/min,检测波长226nm。结果表明,19个药材样品完全符合《中国药典》规定,按干燥品计算,天麻药材中天麻素含量均高于0.1%。冻干处理后,陕西略阳青泥河的天麻含量为所选样本中最优,达到0.624%,而在烘干处理后, 云南昭通彝良县的天麻含量为所选样本中最优,达到0.395%。本实验对天麻药材质量控制做出初步研究,方法快捷、简便,准确度高,重现性好,可以作为天麻药材及其系列制剂的质量控制方法。冻干后所测天麻素含量均高于烘干处理测定值,但各样品在冻干和烘干下的干湿比并无明丝差异,说明烘干条件下干物质的量并无明显损失,天麻素的含量的差异源于自身降解。; Gastrodin in Rhizoma Gastrodiae from Shanxi,Sichuan,Guizhou,Yunnan,Gansu,Anhui, Hunan and Henan province,which were treated with different processing technology,was analyzed and compared by RP-HPLC.The major purpose of the research is to provide the basis for quality control of Rhizoma Gastrodiae and to offer the scientific processing technology for Gastrodin. Shim-pack PREP- ODS(H)KIT C18 column(5μm,250×4.6mm)was used with methanol-phosphate buffer-water(1.5: 3:95.5) as mobile phase.The flow rate was 1.0 mL/min,and the detection wavelength was set at 226 nm.It can be concluded from the results that the content ofgastrodin in all 19 samples ofRhizoma Gastrodiae accorded with the regulations of Chinese Pharmacopoeia,which are higher than 0.1%.After lyophilization,the content of gastrodin in Rhizoma Gastrodiae from Qingnihe,Lueyang,Shanxi province was the highest,which is 0.624%, and after drying,the content of gastrodin in Rhizoma Gastrodiae from Yiliang county,Zhaotong,Yunnan province was the highest,which is 0.395%.This experiment is the preliminary research about the quality control of Gastrodin.The method is swift,simple and convenient,as well as high accuracy and reappearing. The gastrodin content of all the samples treated with lyophilization was higher than those that treated with drying processing technology.However,there is no obvious difference of gastrodin contents processed by different processing technology,so it can be supposed that the self-decomposing of Gastrodin under different temperature is the main reason caused the content difference treated by different processing technology.
