中文摘要目的观察过氧化氢(H2O2)对硫氧还蛋白和APE/Ref-1基因表达的影响,探讨H2O2预处理对神经细胞具有适应性保护的分子机制。方法以PC12细胞作为神经细胞模型,以200和400μmol/L浓度的H2O2处理PC12细胞,用MTT法检测细胞的损伤程度,以Western blot方法测定硫氧还蛋白和APE/Ref-1表达水平。结果200μmol/LH2O2促进PC12细胞的硫氧还蛋白和APE/Ref-1表达,400μmol/LH2O2也促进PC12细胞的APE/Ref-1表达,但使PC12细胞的硫氧还蛋白表达显著降低。结论促进硫氧还蛋白和APE/Ref-1表达可能是低浓度H2O2预处理对神经细胞具有适应性保护的分子机制。; Objective To observe the effect of hydrogen peroxide (H_2O_2) on the expression of thioredoxin and APE/ref-1 and investigate the molecular mechanism of the adaptive protection of H_2O_2 preconditioning.Methods As neural cell model,PC12 cells were treated with 200 and 400 μmol/L of H_2O_2,and the injury of PC12 cells was assessed by MTT.The expression of thioredoxin and APE/ref-1 in PC12 cells was detected by Western blot.Results 200 μmol/L H_2O_2 enhanced the expression of thioredoxin and APE/ref-1 in PC12 cells,and 400 μmol/L H_2O_2 enhanced the expression of APE/ref-1 too.But 400 μmol/L H_2O_2 significantly decreased the expression of thioredoxin.Conclusions The high expression of thioredoxin and APE/ref-1 induced by low concentration H_2O_2 preconditioning may be a part of the molecular mechanism of the adaptive protection of H_2O_2 preconditioning.
