An activatable theranostic prodrug for target cancer therapy and imaging

英文摘要We present a camptothecin–heptamethine cyanine–biotin conjugate (prodrug 8a) that allows for the direct, fluorescence-based monitoring of the targeted cellular uptake and release of a conjugated therapeutic agent. Prodrug 8a is a multifunctional molecule composed of a thiol-specific cleavable disulfide bond, a heptamethine cyanine moiety as a near-infrared fluorescent reporter, camptothecin (CPT) as a model active drug, and biotin as a cancer-targeting unit. Upon addition of free thiols that are relatively abundant in tumor cells, disulfide bond cleavage leads to the tumor-targeted release of the active CPT payload along with near-infrared biotinylated fluorophore 5a. This release is made selective as the result of the biotin functionality. The released nearinfrared biotinylated fluorophore 5a has an emission wavelength significantly different from prodrug 8a, enabling easy detection and monitoring of drug release in vitro and in vivo. Prodrug 8a also would display anticancer activity in the HeLa cell murine xenograft tumor model. The present drug delivery system (DDS) could represent a promising theragnostic strategy, wherein both a therapeutic effect and drug release-related imaging information are produced and can be readily monitored. In due course, prodrug 8a could allow for targeting, more precise monitoring of dosage levels, release, imaging and treatment.
