英文摘要 | Single cell whole transcriptome analysis is a powerful approach for understanding changes in gene expression within an isogenic cell population, and microfluidic device represents a useful tool for high-throughput single-cell analysis. Multilayer microfluidic devices with integrated valves provide an ideal platform for single cell manipulation and analysis, and they have been used extensively in single cell genetic analysis. However, the reaction process of library construction of single cell sequencing is complicated, and using conventional bench top method to do pre-sequencing process is still a common way for many laboratories. Therefore, we aim to develop a useful microfluidic platform to perform single cell extraction with simple operation and high efficiency, and conveniently convert them to bench top experiments. |
