Comparison of Proliferation and Osteogenic Properties of Bone Marrow Mesenchymal Stem Cells between Collagen Induced Arthritis Rat Model and Normal Rat

英文摘要Objective This study aimed to investigate the differentiation of the proliferation and osteogenic properties of bone marrow mesenchymal stem cells(BMSCs) between collagen induced arthritis rat model and normal rat. Methods BMSCs were Isolated, cultured and identified by flow cytometry analysis of CD45 and CD90. The Cell Counting Kit-8 (CCK-8) assay was performed to detect the proliferative activity. The osteogenic properties of bone marrow mesenchymal stem cells between CIA rats and normal ones are analyzed by alizarin red staining,alkaline phosphatase activity and osteocalcin secretion. Results Both kinds of BMSCs were CD90-CD45+ cells. Comparing with normal cells, the proliferation of BMSCs of CIA group are slower, alizarin red staining after osteogenesis induction showed less calcium nodules and coloring shallow, alkaline phosphatase activity and osteocalcin testing showed that the osteogenic capability of BMSCs from CIA group were much weaker as well. Conclusion The osteogenic capability of BMSCs from CIA group decreased significantly,which proved that in addition to the influence of immune cells and active factors, the imbalance of bone absorption and bone formation in RA progress is closely related to BMSCs,or RA itself is a kind of stem cell disease.
