中文摘要基于上转换发光技术生物传感器(Up-Converting Phosphor Technology-Based Biosensor,UPT生物传感器)是一种以上转换发光材料(Up-Converting Phosphor,UCP)作为标一记物的新型免疫检测生物传感器。UCP是由稀土元素掺杂于某些晶体的晶格中构成的纳米级颗粒,它在红外光激发下发射可见磷光。将UCP颗粒作为标记物应用于生物免疫分析中,使UCP生物传感器具有无碎灭、无本底干扰等显著优点,能够获得极高的灵敏度。我们研制的UPT生物传感器用于检测免疫层析试纸条上经生物反应而结合上去的UCP颗粒的含量,继而计算出被测样品中特定生物分子的浓度。本生物传感器以扫描方式工作,由特殊的激光激发与磷光接收系统,信号采集与处理系统及扫描运动系统等三个部分组成。带有被测样品的试纸条被置于扫描平台上,波长为980nln的半导体激光器发出的激光束通过激光激发与磷光接收系统在试纸条表面聚焦为一条极窄的焦线,焦线处的UCP被激发出磷光(波长范围475-670nm)。产生的磷光将被激光激发与磷光接收系统采集,并被光电转换器(如PMT)转换为电信号,该电信号经信号采集与处理系统模一数变换后进入计算机,经数据处理与分析,得到与被测样品相关的信息,如生物反应的阳性或阴性,及被测物的浓度等。论文最后对UPT生物传感器的性能进行了考核。采用20份系列浓度的兔抗鼠疫IgG标准样品考核了UPT生物传感器的检测灵敏度、动态响应特性、稳定性,得到了磷光强度与UPT颗粒浓度的响应曲线;对239份实际血清样品进行了检测与分析,所得结果与WB实验对比检测结果相吻合。
英文摘要The up-converting phosphor technology-based biosensor (UPT-based biosensor) is a novel biosensor in the immunoassay using Up-converting phosphor (UCP) as the biological marker. UCP can emit visible light when excited by infrared light. UPT-based biosensor has many advantages such as no photobleaching and no autofluorecence background. These features provide increased sensitivity and simpler logistical support requirements to the biosensor in bioanalysis. A UPT-based biosensor operating in scanning module has been developed. It includes a specific optical system, a signal control and processing system and a scanning system. A reagent paper with the sample is put on the scanning stage. The laser beam from a 980nm laser diode is focused to a very narrow line on the reagent by the optical system. If the sample contains UCP particles, the particles will emit fluorescence light (475~670nm wavelength) excited by the focal line. The fluorescence can be collect also by the optical system and be converted to electric signal by a photo detector such as a PMT. After specific algorithm using the signal control and processing system, the result can be obtained. To test the performance of the UPT-based biosensor, a series concentration of analytic samples have been used. The UPT system has realized the quantitative detection and had the good ability to meet the need of some emergencies. High sensitivity (nanogram/ml), good linear response characteristics and an excellent correlation (R >0.95) of the UPT-based biosensor have been verified. Some improvement methods have been proposed after the interpretation of result.
