中文摘要毛菊苣(Cichorium glandulosum Boiss. et Huet.)为菊科菊苣属植物,系我国维吾尔医习用药材(毛菊苣维吾尔语为“卡森”)。其药用部分为地上部分、根和种子,性凉,味微苦,具有清热解毒,利水消肿,保肝保肾等功效。现代药理表明,菊苣还具有显著的降糖作用,菊苣提取物明显改善了葡萄糖的吸收及脂类代谢。 为了研究毛菊苣的降糖有效部位,首先将毛菊苣的78个提取部位进行ABTS抗氧化活性测定,并结合体外PTP1B抑制活性及生产成本,确定了一个活性较高的组分进行动物实验,观察了其在体内对糖尿病的治疗效果,发现它不仅具有调节血糖的作用,同时对糖尿病大鼠肾脏也具有一定的保护作用。 其次,进行毛菊苣降糖有效部位的提取工艺研究,通过单因素实验确定了提取溶剂、次数、温度、液比等,并在此基础上采用L9(34)正交试验,以有效部位的粗提率(占30%)以及其对蛋白酪氨酸磷酸酶1B的抑制率(占70%)为指标,考察了溶剂浓度、提取温度、料液比等因素对提取的影响,得到最佳提取工艺条件,并通过扩大验证实验证明此工艺稳定、可行。 再次,进行毛菊苣降糖有效部位纯化工艺研究。以静态吸附法考查9种大孔树脂吸附和解吸附性能及各指标,以吸附率、解吸率、蛋白酪氨酸磷酸酶1B抑制率、ABTS抗氧化性、醛糖还原酶抑制率(各占20%)为指标,进行综合评分,并进行纯化条件的筛选,确定出最佳纯化工艺。
英文摘要Cichorium glandulosum Boiss. et Huet is a plant of Asteraceae chicory genus. As a Uyghur conventional medicine, Cichorium glandulosum plays various roles including detoxification, elimination of swelling and hepato-protective effect. And its medicinal parts are stems, roots and seeds. Modern pharmacological demonstrated that chicory also has a significant hypoglycemic effect, and chicory extract improved the glucose and lipid metabolism. In order to study the hypoglycemic effect of the Cichorium glandulosum, the ABTS antioxidant activity of 78 extract parts from Cichorium glandulosum were determined. Additionally, combined with PTP1B inhibitory activity and production costs, a component with the best activity had been identified for animal experiments. The treatment effect for diabetes was observed and found that it not only regulates the blood glucose, but also have a protective effect on kidney of diabetic rats. The extraction process of the anti-diabetes composition of Cichorium glandulolsum was investigated by one-factor experiment. On the base of which we find how the influence factors (solvent concentration, extraction temperature, the ratio of the solute and solvent, extraction time and extraction times) affect extraction rate and PTP1B inhibition rate. Then a L9(34)orthogonal test was conducted. After calculating a comprehensive score (30% extraction rate add 70% inhibition rate), the optimum extraction process was find. And by expanding the experiment to prove the extraction process is stable and feasible. Finally, the purification of anti-diabetes composition of Cichorium glandulolsum was investigated using macroporous resin. The static adsorption method was to examine the nine kinds of macroporous resin’s adsorption and desorption performance. Adsorption rate, desorption rate, PTP1B inhibition rate, ABTS antioxidant activity and AR inhibition activity were chosen for indicators, and then calculated a composite score ( 20% each ) to determine the best macroporous resin. At last, the purification conditions were screened.
