中文摘要新疆一枝蒿(Artemisia rupestris L.),菊科 (Compositae) 蒿属植物岩蒿的全草,广泛分布于中国新疆,中亚,欧洲等地。一枝蒿是维吾尔医常用药材,具有解毒、健胃消食、抗过敏、抗菌、抗病毒及保肝等功效,主要用于 治疗感冒、咽炎、荨麻诊及肝炎等症。为了更深入的了解新疆一枝蒿药效作用的物质基础,以及更好的开发这一新疆传统药材,本文对该植物的化学成分以及药材 HPLC指纹图谱控制方法分别进行了研究。 对一枝蒿叶中化学成分进行了研究,通过柱层析以及制备HPLC等分离手段,从中分离得到13个生物碱类化合物。并利用现代波谱技术(1H- NMR,13C-NMR,DEPT,1H-1HCOSY,NOESY,ROESY,HMBC,HSQC,ESI-MS,HR-ESI-MS,IR等)对所 分离的生物碱进行了结构鉴定,其中11个为新化合物(1-11)。并对这类化合物的构型和构象进行了研究,发现这类吡啶并环庚烷化合物主要以椅式构象存 在。用泛时密度函理论(TDDFT)对其圆二色谱(CD)进行了模拟计算,结合实验CD谱,确定了此类化合物的绝对构型。 对一枝蒿水溶性部位进行了研究,分离得到了11个化合物,鉴定了其中8个化合物,其中5个为木脂素苷类、2个倍半萜苷类、1个生物碱类化合物。分别为橙皮 素A(citrusin A 14)、 Alaschanioside (15)、 Acorniferin (16)、枸橼苦素(Citrusin B 17) 、丁香树脂酚-4-O-β-D-葡萄糖苷 (Syringaresinol-β-D-glucoside 18)、3-氧代- α-紫罗兰醇-β-D-葡萄糖苷(19)、Byzantionoside B (20)、丁咯地尔(Buflomedil 21),所有化合物均为首次从该植物中发现。 从一枝蒿二氯甲烷部位分离了17个化合物,并鉴定其中了11个化合物,分别为金腰素乙 (Chrysosplenetin B 22)、洋艾素 (Artemetin 23)、金合欢素 (Acacetin 24)、3,5,4'-三羟基-6,7,3'-三甲氧基黄酮 (3,5,4'-Trihydroxy-6,7,3'-trimethoxyflavone 25)、雷杜辛黄酮醇 (Retusine C 26)、槲皮素-3, 7, 3' -三甲醚 (Pachypadol 27)、异山奈甲黄素(Isokaemapferide 28)、酮酸 (Rupestonic Acid 29)、7-甲氧基香豆素 (hermiarin 30)、 β-谷甾醇 (β-sitosterol 31)、二十四烷基-1,3-二醇(tetracosane-1,3-diol 32),其中化合物3-7为首次从本植物中分离得到。 最后研究了一枝蒿HPLC指纹图谱的质量控制方法,通过对14批不同产地一枝蒿药材的研究建立了一枝蒿药材的指纹图谱。其中14个色谱峰为共有峰。利用 HPLC指纹图谱相似性评价软件评价,10批药材的相似度在0.991~0.678之间。
英文摘要Artemisia rupestris L.(Compositae) is widely distributed in the Xinjiang Uighur autonomous region of China, central Asia, Europe, etc. It is a well-known traditional Chinese medicinal plant in Xinjiang and pharmacological studies showed its activities of clearing away toxins, strengthening the stomach to promote digestion, anti-hypersusceptibility, antibacteria, antivirus and protecting liver. Clinically, it si frequently used to treat cold, pharyngitis, urticaria, hepatitis and etc. In order to find out the active compoenets, and to further develop the traditional Uyghur medicinal material, we studied on the chemical constituents and HPLC fingerprints of Artemisia rupestris L. To study the chemial constitents of leaves of Artemisia rupestris L. Thirteen alkaloids were isolated by means of column chromatography and preparative HPLC. The structure of these compounds were elucidated by spectroscopic methods: 1H-NMR, 13C-NMR, DEPT, 1H-1HCOSY, NOESY, ROESY, HMBC, HSQC, ESI-MS, HR-ESI-MS, IR. Among them, eleven compounds are new. According to coupling constant and conformational analysis by molecular mechanics calculation, all compounds existed mainly in the chair conformation with the substituent at C-8 in the equatorial position. The absolute configuration of them was assigned using NMR, CD data and TDDFT calculations. To investigate the chemical constituents of water-soluble fraction from Artemisia rupestris L. 7 known glycosides were isolated and identigied as acitrusin A (14), alaschanioside (15), acorniferin (16), citrusin B( 17), syringaresinol-β-D-glucoside (18), 3-oxo- α-ionol- glucopyranoside (19)、byzantionoside B (20). In addition, an alkaloid called Buflomedil (21) was obtained. All of them were isolated from this plant for the first time. To investigate the chemical constituents of anti-virus fraction from Artemisia rupestris L. Eleventeen copounds were isolated and their sturctures were identified as chrysosplenetin B (22), artemetin (23), acacetin (24), 3,5,4'-Trihydroxy-6,7,3'-trimethox- yflavone(25), retusine C (26), pacphypadol (27), isokaemapferide(28), rupestonic acid (29), hermiarin (30), β-sitosterol(31), tetracosane-1,3-diol(32). Among them , comounds (24-27) were obtained for the first time . To study the determination method of Artemisia rupestris L.. The fingerprint of Artemisia rupestris L. with 14 common peaks was established by study of 10 different sources. Estimation the fingerprint by the similarity analysis of HPLC fingerprints spectra.The result of comparability was between 0.99185~0.678.
